Research in basic science is supported by experimental data one by one, and the same is true for exploration in life sciences. The generation of every experimental data is inseparable from the contribution and dedication of experimental animals, and they are all precious.

On the morning of August 28, 2021, the Cryomedicine Research Center of Shandong Yinfeng Life Sciences Research Institute, the research unit of our foundation, welcomed two special "volunteers": female beagles named Cryobig8 and Cryobig9 respectively. The eighth and ninth animal "volunteers" to participate in the experiment since the launch of the Yinfeng Life Extension Project. In order to more accurately and intuitively monitor key technical indicators such as temperature, pressure, flow rate and protective agent replacement degree during cryopreservation surgery and perfusion, as well as the operation of the cooling program, Cryobig8 and Cryobig9 will be verified through different cryopreservation experimental surgeries.

 

Use experiments to verify the scientific nature of surgery

At 10:00 a.m., cryogenic surgery officially began. After Cryobig8 was anesthetized, surgical experts established an extracorporeal circulation-connected cryogenic surgical perfusion detection system for him, turned on the extracorporeal circulation machine and membrane lung, and quickly lowered his body temperature to below 18°C through a low-temperature circulating water bath, and quickly performed basic perfusion fluid and Cyclic perfusion of cryoprotectants with different concentrations. As a control experiment, Cryobig9's perfusion method was different, using only its own blood for low-temperature circulation cooling.

After the perfusion was completed, the experimental dogs were transferred to a programmed cooling device that was pre-cooled to 4°C. The programmed cooling was started. After more than 20 hours, the temperature was reduced to -190°C, and then transferred to an independent stainless steel liquid nitrogen storage tank for liquid phase storage.

The surgery and perfusion lasted from 10 am to 17 pm on the 4th, which took 7 hours. A team of clinical experts including cardiac surgery experts Sun Wenyu and Zhao Xin, anesthesia experts Hou Yuedong and Li Huihui, and extracorporeal circulation specialist Zhao Fen participated in the demonstration and implementation of the experimental plan.

In order to verify the protective effect of the cryoprotective solution on the body and tissues and organs, after the perfusion was completed, some kidneys, livers and muscle tissues from different parts of the experimental period were taken as samples, and the frozen samples were used to analyze tissue cell morphology, activity, etc. detection.

According to the predetermined experimental plan, the two experimental dogs will also undergo brain CT scans in the future to compare and analyze the perfusion effect of cryoprotectant and the structural changes of tissues and organs after preservation.
